Abstract. In this study, a simple, rapid and sensitive kinetic sepectrophotometric method for trace amounts determination of vitamin B12 was developed. Orange-G was oxidized by bromate in acidic media and vitamin B12 has inhibitory effect on it. The reaction was followed spectrophotometrically at 478 nm (λmax). Under optimum experimental conditions (H2SO4, 0.84 mol L-1; Orange-G, 66.0 µmol L-1; bromat, 5.0 mmol L-1; 20 oC and 360 s), vitamin B12 can be determined in concentration range 0.1 – 2.2 µg mL-1 and the detection of limit was 0.02 µg mL-1 of vitamin B12. The proposed method were successfully applied for the determination of vitamin B12 in human serum as biological and pharmaceutical factory as real sample and satisfactory results were obtained.
